Review: The value of Drosophila in VUS resolution and classification

We discuss how different experimental paradigms and genetic tools in Drosophila can be used to end the diagnostic odyssey of people living with undiagnosed diseases. We further argue that flies can help classify pathogenic variants into different allelic classes through genetic experiments, and this information can help develop personalized therapeutic strategies.
